Immune Repertoire Analysis

The adaptive immune receptor repertoire (AIRR) represents the complete collection of all B-cell receptor (BCR) and T-cell receptor (TCR) sequences within an individual at a given time point. It serves both as the effector of adaptive immune responses and as a "molecular recorder" of the body's health and disease status. Its diversity originates from stochastic V(D)J segment rearrangement and random nucleotide additions and deletions at the junctions. Among all regions, the complementarity-determining region 3 (CDR3) exhibits the greatest variability and most critically determines antigen-recognition specificity, making it the central focus of repertoire analysis. ZhuHai GeneRulor has established a comprehensive immune repertoire sequencing and analysis platform covering both bulk and single-cell levels for TCR and BCR, in accordance with internationally recognized methodological reviews and industry standards, including NGS-based immunoglobulin/T-cell receptor rearrangement detection and the EuroClonality-NGS clonality assessment framework. The platform is structured around three key dimensions — starting template (gDNA/RNA), library preparation strategy (multiplex PCR/5'RACE), and single-cell chain pairing — and offers six major product lines: TCR/BCR multiplex PCR sequencing (FFPE-compatible, with cell-equivalent quantification), TCR/BCR 5'RACE full-length sequencing (incorporating UMI-based error correction for unbiased full-length V(D)J recovery), and single-cell TCR/BCR sequencing (native α–β / VH–VL chain pairing, integrable with single-cell transcriptomics). These are complemented by a systematic bioinformatics analysis suite encompassing clonotype identification and abundance profiling, diversity and clonality indices, V/D/J gene usage and CDR3 characterization, clonal lineage tracing, somatic hypermutation (SHM) analysis, and machine learning-based modeling.

The platform serves a broad range of applications, including clonal dynamics analysis in autoimmune and infectious diseases, tumor immune response and tumor-infiltrating lymphocyte (TIL) clone-by-phenotype studies, vaccine immunodynamics and antibody affinity maturation monitoring, antibody and TCR therapeutic candidate discovery, as well as clonal tracking and mechanistic studies for CAR-T/TCR-T cell therapies. Leveraging GeneRulor's deep expertise in gene editing, the platform provides an integrated one-stop monitoring solution for cell therapy products — combining off-target detection, integration site analysis, and immune repertoire profiling (including single-cell) — delivering a synergistic technical advantage unattainable by pure sequencing services alone. The platform delivers raw FASTQ data along with transparent, reproducible analysis pipelines, supporting multiple species including human and mouse, and accommodating diverse sample types such as blood, tissue, FFPE, and viable cells. It provides a professional and reliable end-to-end immune repertoire solution for basic research, translational medicine, and drug development.
